    Development of the City Public Service Model on the Basis of Integrated Transport Flow Indicators

    The problem of modeling public services based on architectural and planning decisions is considered, the role of traffic in the formation of a model of city services is analyzed. An integrated criterion for the quality of public services is proposed. A method has been developed for determining segmented public services taking into account the transport areas of the city, which will make it possible to evenly disperse public service centers. The basis is a socio-planning organization, as a material-spatial system containing anthropogenic and natural components – the territory and institutions where the functional processes that take place in the urban planning environment take place. The described model has certain versatility, and is simultaneously suitable for characterizing various categories of service institutions. Thus, the task of optimizing the quality of public services in the city is reduced to a mathematical model for which, by setting the basic design criteria, the optimal result can be obtained.On the basis of a questionnaire survey and analysis of statistical data, calculation of traffic intensity, the demand and supply of the level of public services Ρ„re studied. The structural elements of this model: population, territory, transport and service institutions, are in dialectical interaction, which is described by the mathematical model in this study. The model is based on the calculation of the minimum population in the service area, which allows to have i-th type establishments using the social potential method, as well as a graph-analytical method in determining the optimal location of service institutions in the city.As a result of the research, a conceptual model of public services for cities is built, a layout of supermarkets in the territory of Uzhhorod and distribution of service areas of these institutions is proposed. This optimization will ensure uniform domestic servicing of the territory, optimal performance indicators of service establishments and minimum average service radii of points

    Oxygen and Carbon Dioxide Rhythms Are Circadian Clock Controlled and Differentially Directed by Behavioral Signals

The final version is available from Elsevier (Cell Press).Daily rhythms in animal physiology are driven by endogenous circadian clocks in part through rest-activity and feeding-fasting cycles. Here, we examined principles that govern daily respiration. We monitored oxygen consumption and carbon dioxide release, as well as tissue oxygenation in freely moving animals to specifically dissect the role of circadian clocks and feeding time on daily respiration. We found that daily rhythms in oxygen and carbon dioxide are clock controlled and that time-restricted feeding restores their rhythmicity in clock-deficient mice. Remarkably, day-time feeding dissociated oxygen rhythms from carbon dioxide oscillations, whereby oxygen followed activity, and carbon dioxide was shifted and aligned with food intake. In addition, changes in carbon dioxide levels altered clock gene expression and phase shifted the clock. Collectively, our findings indicate that oxygen and carbon dioxide rhythms are clock controlled and feeding regulated and support a potential role for carbon dioxide in phase resetting peripheral clocks upon feeding.British Heart FoundationEuropean Research CouncilEuropean Union, Seventh Framework Program, Marie Curie Action

    Should I keep studying? Consequences of a decision to stop learning in young and older adults

    In situations of cognitive overload, the role of a metacognitive decision to stop learning is of utmost importance. We investigated how young and older adults decide to stop learning as a strategy for maximizing memory performance when they face to-be-learned material exceeding their memory capability. People may decide to stop learning for two main reasons: they experience a growing feeling of disfluency as a learning episode progresses and/or they perceive such a decision to be beneficial for future memory performance. In Experiments 1 and 2, participants studied lists of 50 words. The majority of young and older adults stopped learning in conditions where they were allowed to do so. This decision, counterintuitively, decreased the number of recalled words. Crucially, a similar number of young and older adults stopped the presentation of to-be-remembered material, and both age groups suffered comparable consequences in their memory performance. In Experiments 3a and 3b, participants read an experimental scenario and decided whether they would stop learning based on this description alone. People in different age groups predicted their metacognitive decisions similarly. However, participants’ forecasted performance did not reflect the negative influence of these decisions. Regardless of their age, people tend to make a suboptimal decision to stop learning, unaware of its negative consequences. Together, our results suggest that young and older adults can exert metamemory control to similar degrees even though their decisions may not be beneficial for memory performance.     Multilingual representations for low resource speech recognition and keyword search

This paper examines the impact of multilingual (ML) acoustic representations on Automatic Speech Recognition (ASR) and keyword search (KWS) for low resource languages in the context of the OpenKWS15 evaluation of the IARPA Babel program. The task is to develop Swahili ASR and KWS systems within two weeks using as little as 3 hours of transcribed data. Multilingual acoustic representations proved to be crucial for building these systems under strict time constraints. The paper discusses several key insights on how these representations are derived and used. First, we present a data sampling strategy that can speed up the training of multilingual representations without appreciable loss in ASR performance. Second, we show that fusion of diverse multilingual representations developed at different LORELEI sites yields substantial ASR and KWS gains. Speaker adaptation and data augmentation of these representations improves both ASR and KWS performance (up to 8.7% relative). Third, incorporating un-transcribed data through semi-supervised learning, improves WER and KWS performance. Finally, we show that these multilingual representations significantly improve ASR and KWS performance (relative 9% for WER and 5% for MTWV) even when forty hours of transcribed audio in the target language is available.     In the recently characterized Type IX Secretion System (T9SS), the conserved C-terminal domain (CTD) in secreted proteins functions as an outer membrane translocation signal for export of virulence factors to the cell surface in the Gram-negative Bacteroidetes phylum. In the periodontal pathogen Porphyromonas gingivalis, the CTD is cleaved off by PorU sortase in a sequence-independent manner, and anionic lipopolysaccharide (A-LPS) is attached to many translocated proteins, thus anchoring them to the bacterial surface. Here, we solved the atomic structure of the CTD of gingipain B (RgpB) from P. gingivalis, alone and together with a preceding immunoglobulin-superfamily domain (IgSF). The CTD was found to possess a typical Ig-like fold encompassing seven antiparallel Ξ²-strands organized in two Ξ²-sheets, packed into a Ξ²-sandwich structure that can spontaneously dimerise through C-terminal strand swapping. Small angle X-ray scattering (SAXS) revealed no fixed orientation of the CTD with respect to the IgSF. By introducing insertion or substitution of residues within the inter-domain linker in the native protein, we were able to show that despite the region being unstructured, it nevertheless is resistant to general proteolysis. These data suggest structural motifs located in the two adjacent Ig-like domains dictate the processing of CTDs by the T9SS secretion pathway

    Whole genome resequencing of black Angus and Holstein cattle for SNP and CNV discovery

    Background: One of the goals of livestock genomics research is to identify the genetic differences responsible for variation in phenotypic traits, particularly those of economic importance. Characterizing the genetic variation in livestock species is an important step towards linking genes or genomic regions with phenotypes. The completion of the bovine genome sequence and recent advances in DNA sequencing technology allow for in-depth characterization of the genetic variations present in cattle. Here we describe the whole-genome resequencing of two Bos taurus bulls from distinct breeds for the purpose of identifying and annotating novel forms of genetic variation in cattle.Results: The genomes of a Black Angus bull and a Holstein bull were sequenced to 22-fold and 19-fold coverage, respectively, using the ABI SOLiD system. Comparisons of the sequences with the Btau4.0 reference assembly yielded 7 million single nucleotide polymorphisms (SNPs), 24% of which were identified in both animals. Of the total SNPs found in Holstein, Black Angus, and in both animals, 81%, 81%, and 75% respectively are novel. In-depth annotations of the data identified more than 16 thousand distinct non-synonymous SNPs (85% novel) between the two datasets. Alignments between the SNP-altered proteins and orthologues from numerous species indicate that many of the SNPs alter well-conserved amino acids. Several SNPs predicted to create or remove stop codons were also found. A comparison between the sequencing SNPs and genotyping results from the BovineHD high-density genotyping chip indicates a detection rate of 91% for homozygous SNPs and 81% for heterozygous SNPs. The false positive rate is estimated to be about 2% for both the Black Angus and Holstein SNP sets, based on follow-up genotyping of 422 and 427 SNPs, respectively. Comparisons of read depth between the two bulls along the reference assembly identified 790 putative copy-number variations (CNVs). Ten randomly selected CNVs, five genic and five non-genic, were successfully validated using quantitative real-time PCR. The CNVs are enriched for immune system genes and include genes that may contribute to lactation capacity. The majority of the CNVs (69%) were detected as regions with higher abundance in the Holstein bull.Conclusions: Substantial genetic differences exist between the Black Angus and Holstein animals sequenced in this work and the Hereford reference sequence, and some of this variation is predicted to affect evolutionarily conserved amino acids or gene copy number. The deeply annotated SNPs and CNVs identified in this resequencing study can serve as useful genetic tools, and as candidates in searches for phenotype-altering DNA differences

    Biosynthesis and Molecular Genetics of Polyketides in Marine Dinoflagellates

    Marine dinoflagellates are the single most important group of algae that produce toxins, which have a global impact on human activities. The toxins are chemically diverse, and include macrolides, cyclic polyethers, spirolides and purine alkaloids. Whereas there is a multitude of studies describing the pharmacology of these toxins, there is limited or no knowledge regarding the biochemistry and molecular genetics involved in their biosynthesis. Recently, however, exciting advances have been made. Expressed sequence tag sequencing studies have revealed important insights into the transcriptomes of dinoflagellates, whereas other studies have implicated polyketide synthase genes in the biosynthesis of cyclic polyether toxins, and the molecular genetic basis for the biosynthesis of paralytic shellfish toxins has been elucidated in cyanobacteria. This review summarises the recent progress that has been made regarding the unusual genomes of dinoflagellates, the biosynthesis and molecular genetics of dinoflagellate toxins. In addition, the evolution of these metabolic pathways will be discussed, and an outlook for future research and possible applications is provided
